This research aims to develop an effective diagnostic biomarker to identify breast cancer using ellagic acid compound found in pomegranate (Punica granatum) as a specific target. The research method was conducted using Pymol, Pyrex, Protein Plus, and Lepinski Rule software for structural analysis and molecular interactions. The results showed that ellagic acid has a strong binding affinity for the protein-tyrosine kinase receptor ERBB-2, with binding affinity values of -6.7, -6.6, and -6.5 and RMSD of 0, 0.147, and 1.301. In addition, analysis using Protein Plus revealed an interaction between ellagic acid and the protein-tyrosine kinase receptor ERBB-2. Lipinski analysis showed that ellagic acid has a mass of 302, a hydrogen bond donor number of 4, a hydrogen bond acceptor number of 8, a log P of 1.241, and a molar reactivity of 68.454. This discovery has the potential to be an effective diagnostic biomarker in identifying breast cancer, which could help in the early diagnosis and treatment of this disease.

Introduction: The footprint of Neuregulin 1 (NRG1) / ERbB4 in the pathophysiology of some neurological disorders and TRPV1 regulation has been indicated. The alterations in NRG1 and ErbB4 as well as the TRPV1 signaling pathway were investigated during the development of absence epilepsy in the genetic animal model of absence epilepsy. Methods: Male WAG/Rij and Wistar rats were divided into four experimental groups of two and six months of age. The protein levels of NRG1, ERbB4, and TRPV1 were measured in the somatosensory cortex and hippocampus. Results: The cortical protein levels of NRG1 and ErbB4 in the 6-month-old WAG/Rij rats were lower than in Wistar rats. Protein levels of TRPV1 were lower in two-and sixmonth-old WAG/Rij rats compared to age-matched Wistar rats. Hippocampal protein levels of NRG1 in 6-month-old WAG/Rij rats were lower than two-month-old WAG/Rij rats. Low levels of ErbB4 protein in two-month-old and high levels in six-month-old WAG/Rij rats were found compared to Wistar rats. Protein levels of TRPV1 were lower in the two-month-old and higher in the six-month-old WAG/Rij rats compared to age-matched Wistar rats. Furthermore, a high correlation between NRG1/ERbB4 and TRPV1 expressions in the cortex and hippocampus was indicated. The expression of NRG1/ERbB4 and TRPV1 followed a similar pattern during the life span of Wistar and WAG/Rij rats. Conclusion: Our findings indicated the potential role of the NRG1/ErbB4 pathway as well as TRPV1 in the pathogenesis of absence epilepsy. The regulatory effect of the ERbB4 receptor on the TRPV1 expression has been suggested following the similar pattern of expression.

The emerging era of personalized medicine makes it increasingly important toconsider intratumoral heterogeneity, which has been found in some breast cancercases. However, its identification criteria, form of reporting, and subsequent effects onthe clinical course of this disease remain controversial and not fully defined. Here, wereport and discuss a case of breast invasive ductal adenocarcinoma with substantialintratumoral heterogeneity, discrepancy between Her2/neu immunostaining and insitu hybridization, and disparity between estrogen receptor status before and afterneoadjuvant therapy.

Background: Ovarian cancer is the second most prevalent gynecological malignancy. Its prognosis is poor with a five-year survival rate of <50% with the available therapies. There is a constant need for new biological markers. Therefore, we conducted the present study to evaluate the association between human epidermal growth factor receptor (HER2) neu and the clinicopathological features of epithelial ovarian cancer.Method: A prospective, observational analytic study was conducted at Fauji Foundation Hospital, Rawalpindi between November 2018 and October 2019. It was a cross-sectional study with a quantitative correlational study design.Results: We recruited 90 patients. The mean age of diagnosis was 53 ± 8.022 years; 81.1% (n = 73) had raised pretreatment CA-125 levels with stage III (56.7%, n = 51) and grade III (54.4%, n = 49) at presentation. It was seen that 24.4% (n = 22) of the tumors expressed HER2/neu, 65.6% (n = 59) were negative and 10% (n = 9) were equivocal. 72.2% had platinum sensitive disease. According to HER2/neu status, 20% HER2/neu positive patients had platinum sensitive disease and 3.3% had resistant disease; meanwhile, 47.8% HER2/neu negative patients had platinum sensitive disease and 10% were platinum resistant. HER2/neu expression was significantly associated with grade (P = 0.040) and pretreatment CA-125 levels (P = 0.032) whereas our study failed to show a significant association between stage (P = 0.383) and chemotherapy response (P = 0.055).Conclusion: The current study demonstrated that HER2/neu was positive in 24.4% of the patients, which was significantly associated with grade and pretreatment CA-125 level. However, a longer follow-up is needed for survival analysis and establishment of HER2/neu as a prognostic marker for epithelial ovarian cancer.
